Before any therapeutic effect can occur, the administered agent must physically reach the tumor, a process influenced by factors such as blood supply to the tumor, vascular permeability, and the structural barriers presented by the surrounding tissue.

Achieving Sufficient Local Concentration

Reaching the tumor site alone is not sufficient; the treatment must accumulate at a concentration high enough to produce a meaningful biological effect, since exposure below this threshold may have negligible impact regardless of the agent's inherent potency.
