The primitive heart tube is composed of several contiguous domains, each with unique molecular identities and developmental fates.


Principal Regions of the Primitive Heart Tube

Sinus Venosus Region

The sinus venosus is the most caudal (inflow) segment of the primitive heart tube. It receives venous blood from the embryo, yolk sac, and placenta via the common cardinal, vitelline, and umbilical veins. The sinus venosus contributes to the formation of the smooth-walled part of the right atrium (sinus venarum), the coronary sinus, and the oblique vein of the left atrium.

Primitive Atrium Region

Anterior to the sinus venosus, the primitive atrium forms a single chamber that later partitions into right and left atria. The primitive atrium gives rise primarily to the auricles (pectinate-walled parts) of both adult atria.

Primitive Ventricle Region

The primitive ventricle lies just cranial to the primitive atrium and primarily forms the trabeculated region of the adult left ventricle. Its muscular wall is the source of the bulk of the future left ventricular myocardium.

Bulbus Cordis Region

The bulbus cordis is positioned cranial to the primitive ventricle. It consists of three subregions: the proximal bulbus (forms most of the right ventricle), the conus cordis (outflow tracts of both ventricles), and the distal truncus arteriosus (future roots of the great arteries).

Conus Cordis Region

The conus cordis is the intermediate segment between the bulbus cordis and truncus arteriosus. It forms the outflow tracts that will eventually become the infundibulum of the right ventricle and the aortic vestibule of the left ventricle.

Truncus Arteriosus Region

The truncus arteriosus is the most cranial (outflow) part of the heart tube and connects directly with the aortic sac. It gives rise to the proximal portions of the ascending aorta and pulmonary trunk.

Aortic Sac Region

The aortic sac is the distal outflow chamber of the heart tube, from which the aortic arches arise. These arches connect to the dorsal aorta and are essential for patterning the major arteries of the head and neck.


Inflow-to-Outflow Tube Axis

The primitive heart tube is organized along a cranio-caudal axis, with the sinus venosus at the inflow (caudal) end and the aortic sac at the outflow (cranial) end. Blood flows sequentially from the sinus venosus → primitive atrium → primitive ventricle → bulbus cordis → conus cordis → truncus arteriosus → aortic sac.
